*** in RH linux , after compiling your kernel with ipv6 enabled options , you will have to run ifconfig command as

ifconfig eth0 add anyaddr 



where anyaddr means the ipv6 address u want to assign.

after giving the address u can test with ping command.

*** the prefix you will set in the translator will be given to the ipv4 address , so any prefix will do .
u also need to set in ur h1 machine to route anything destined to prefix:ipv4address to route to your h2 machine.

for that u will have to use route command.

*** here ping & most ipv6 compliant tools will work ,
i think the translator program also includes dns_alg[dns application level gateway] which can also run convert ur dns queries, so nslookup will also work , but u need to set nameserver.
i think  translator also includes ftp-alg , so using that program ftp will also work.
